attachment

ever since monday, i have been going to "skl" at NTU, school of biological sciences. yep thats right. all the way at BOON LAY when i stay in marine parade. my goodness. so anyhow, allister, marcus and i are under the same prof for the attachment...so on the first day, morning, all of the NTU attachment ppl went to skl to report and then took a bus to NTU. after waiting in this vast lobby for i dunno HOW long, Dr andrew tan finally came and he gave us a tour of the humongous labyrinth of a school. showing us all the labs, pantries, computer room, meeting room, cafe and such...its REALLLLLLLLY easy to get lost in that place....after we all walked arnd the place for a couple of hours, we walked all teh way to the canteen which is a couple of schools away....for lunch. my god..that place was PACKED...the ridiculous thing is...we went to lunch at 1130. the teachers told us to meet back at the building at TWO pm. my GOODNESS. we had like SO MUCH TIME to kill. oh yah....i forgot to mention the numerous number of locked doors that requires a card to access. so SEVERAL hours after lunch, we finally got to meet our prof...who is a reallly cute looking grandfather type man...complete with a big belly and balding hair. but he seems a really nice person lah...humorous too...not to mention freaking smart...so that day we basically did nothing but tok to him....and yah. went home late...one hour and a half of travelling...my GOD. anyway, on the second day, we had a safety briefing by another funny prof and when we got our lab coats, we started to work. well, most of the day we were waiting for our sorta mentor who told us the overview of wad we were gonna do, showed us how to do PCR by mixing all the primers and dna and whatnot and then just put into the PCR machine......and bringing us arnd the skl to look at the equipments like the autoclaving machine and oven and ice machine. so late in the evening, we finally started our lab work...which was actually nothing much..just cleaned the clean hood with ethanol spray and pipetting LB broth into 3 universals and then using a toothpick, we just scraped off a colony of bacteria and dumped it into the broth and left it in the incubator which rotates constantly...and hen we went home after that....so we waited one whole day to scrape that colony off the agar plate.. today, we reported at 10am...after reading about minipreps, we went to the lab to do that on thebacteria culture that we sorta grew the day before. but first we made this glycerol stock...which was just like..adding glycerol to the eppendorf tube and vortexing it and then freezing it....so following the instructions, we added buffers, vortexing, add another buffer and mix by inverting...then after that add yet another buffer and then centrifuge it. then we took out the supernatant and put it in some spin column and centrifuge it, pour the flowthrough and then put ANOTHER buffer and spin again and pour away the flowthrough and then move part of the spin colomn with the plasmid that we wanna isolate to a eppendorf tube and add water, let it stand for a min then spin again...and finally managed to isolate the plasmid...and after lunch, kitwei showed us how to see the density of the DNA...yah...and after that we went home. hmmmmm...i like the tap cards....its kewl...:P
